摘要: Assisted reproduction provides a wide spectrum of treatments and strategies addressing infertility. However, distinct groups of infertile patients with unexplained infertility, congenital disorders, and other complex cases pose a challenge in fertilization (IVF) practices. This special cohort of patients is associated with futile attempts, IVF overuse, and dead ends in management. Cutting edge research on animal models introduced this concept, along with the development of artificial organs with the aim to mimic the respective physiological functions in reproduction. Extrapolation on clinical application leads to the future use of infertility management in humans. To date, the successful clinical application of artificial reproductive organs in humans is not feasible because further animal model studies are required prior to clinical trials. The application of these artificial organs could provide a solution to infertility cases with no other options. This manuscript presents an overview on the current status, future prospects, and considerations on the potential clinical application of artificial ovary, uterus, and gametes in humans. This paper presents how the IVF practice landscape may be shaped and challenged in the future, along with the subsequent concerns in assisted reproductive treatments.

摘要: Polycystic ovary syndrome (PCOS) is a common gynecologic endocrinopathy. The pathogenesis of PCOS is associated with both heredity and environment. PCOS has adverse impacts on female endocrine, reproduction, and metabolism. PCOS can impact women’s reproductive health, leading to anovulatory infertility and higher rate of early pregnancy loss. PCOS has additional metabolic derangements, such as insulin resistance, impaired glucose tolerance, and dyslipidemia. The risks of diabetes, cardiovascular disease, hypertension, metabolic syndrome, and endometrial cancer among PCOS patients are significantly increased as well.

The activity of free radicals in follicular fluid was related to ovarian responsiveness, in vitro fertilization (IVF), and embryo transfer success rate. However, studies analyzing the relationship between the free radical scavenging capacity and embryo quality of infertile women with polycystic ovarian syndrome (PCOS) were lacking. The aim of this study was to evaluate the relationship between the free radical scavenging window of women with PCOS and their embryo quality. The free radical scavenging capacity of follicular fluid from women with PCOS was determined by a,a-diphenyl-b-picrylhydrazyl (DPPH), 2,2-azinobis (3-ethylbenzthiazoline-6-sulphonic acid) assay, superoxide radical, and reactive oxygen species (ROS) assay. In the DPPH and ROS assays, the follicular fluid from grades I and II embryos was significantly higher than the follicular fluid from grades III and IV embryos. The lower control limit of DPPH radical scavenging capacity and upper control limit of ROS level were 13.2% and 109.0 cps, respectively. The calculated lower control limit and upper control limit were further confirmed in the follicular fluid of embryos of all grades. These cut-off values of free radical scavenging activity of follicular fluid could assist embryologists in choosing the development of embryos in PCOS patients undergoing IVF.

The increased levels of intracellular reactive oxygen species (ROS) in granulosa cells (GCs) may affect the pregnancy results in women with polycystic ovary syndrome (PCOS). In this study, we compared thein vitro fertilization and embryo transfer (IVF-ET) results of 22 patients with PCOS and 25 patients with tubal factor infertility and detected the ROS levels in the GCs of these two groups. Results showed that the PCOS group had significantly larger follicles on the administration day for human chorionic gonadotropin than the tubal factor group (P<0.05); however, the number of retrieved oocytes was not significantly different between the two groups (P>0.05). PCOS group had slightly lower fertilization, cleavage, grade I/II embryo, clinical pregnancy, and implantation rates and higher miscarriage rate than the tubal factor group (P>0.05). We further found a significantly higher ROS level of GCs in the PCOS group than in the tubal factor group (P<0.05). The increased ROS levels in GCs caused GC apoptosis, whereas NADPH oxidase 2 (NOX2) specific inhibitors (diphenyleneiodonium and apocynin) significantly reduced the ROS production in the PCOS group. In conclusion, the increased ROS expression levels in PCOS GCs greatly induced cell apoptosis, which further affected the oocyte quality and reduced the positive IVF-ET pregnancy results of women with PCOS. NADPH oxidase pathway may be involved in the mechanism of ROS production in GCs of women with PCOS.

美洲鲥鱼卵巢发育规律和性类固醇激素变化研究

摘要:

采用组织学、形态学及电化学等技术方法,研究了人工养殖条件下美洲鲥鱼雌性亲鱼卵巢发育和性类固醇激素的周期变化规律。结果表明,美洲鲥鱼属于非同步分批产卵类型;根据卵巢外部形状大小及色泽等的不同,可将美洲鲥鱼卵巢发育过程分为6 个时期;根据卵母细胞的形态结构、卵黄物质的积累和滤泡细胞的变化,将卵子发生划分为6 个时相;卵巢周期变化过程中,肝脏指数(HSI)和肥满度(CF)变化不显著,性腺指数(GSI)与血浆中雌二醇(E2)水平表现出与性腺发育协同一致的变化规律。

遗传因素、生活方式和环境因素与多囊卵巢综合征(PCOS)的发生相关。身体质量指数(BMI)的增加会加剧生殖及代谢障碍,降低PCOS女性的生育能力。本文对在中国大陆21个研究中心开展的一项大样本、多中心、随机对照试验进行二次分析。共有1000名PCOS女性参加本试验。其中,998名PCOS女性被纳入分析。BMI增加与月经失调加剧、高睾酮水平、高代谢综合征患病率以及生活质量降低相关。BMI正常组、超重组和肥胖组的女性人均排卵率分别为83.0%、78.2%和63.6%(P < 0.001),活产率分别为23.6%、18.1%和15.3%(P = 0.030)。与中国南方的PCOS患者相比,北方的PCOS患者生殖结局较差,血糖和血脂代谢不良,运动量少,以及总排卵率更低(81.2% vs 74.8%,绝对差:6.4%,95%置信区间:1.2%~11.5%)。结果显示,中国南北方的汉族PCOS女性具有典型的表型特征。与生活在中国南方的女性相比,在同样的干预措施下,中国北方的女性BMI更高,糖脂代谢障碍更严重,临床结局更差。表型特征及排卵的差异可以通过BMI差异加以解释。

Applications of traditional pump design theory to artificial heart and CFD simulation

摘要: A novel heart pump model was obtained by improving the traditional axial pump design theory with the consideration of working and hydraulic situations for artificial hearts. The pump head range and the velocity triangle were introduced and an iterative approach was utilized for the initial model. Moreover, computational fluid dynamics (CFD) simulations were performed to determine relevant model parameters. The results show that this procedure can be used for designing a series of high-efficiency artificial heart pumps.

INTERACTIVE KNOWLEDGE LEARNING BY ARTIFICIAL INTELLIGENCE FOR SMALLHOLDERS

Enhancement of farming management relies heavily on enhancing farmer knowledge. In the past, both the direct learning approach and the personnel extension system for improving fertilization practices of smallholders has proven insufficiently effective. Therefore, this article proposes an interactive knowledge learning approach using artificial intelligence as a promising alternative. The system consists of two parts. The first is a dialog interface that accepts information from farmers about their current farming practices. The second part is an intelligent decision system, which categorizes the information provided by farmers in two categories. The first consists of on-farm constraints, such as fertilizer resources, split application times and seasons. The second comprises knowledge-based practices by farmers, such as nutrient in- and output balance, ratios of different nutrients and the ratios of each split nutrient amount to the total nutrient input. The interactive knowledge learning approach aims to identify and rectify incorrect practices in the knowledge-based category while considering the farmer’s available finance, labor, and fertilizer resources. Investigations show that the interactive knowledge learning approach can make a strong contribution to prevention of the overuse of nitrogen and phosphorus fertilizers, and mitigating agricultural non-point source pollution.

Artificial intelligence in gastroenterology: where are we heading?

摘要: Artificial intelligence (AI) is coming to medicine in a big wave. From making diagnosis in various medical conditions, following the latest advancements in scientific literature, suggesting appropriate therapies, to predicting prognosis and outcome of diseases and conditions, AI is offering unprecedented possibilities to improve care for patients. Gastroenterology is a field that AI can make a significant impact. This is partly because the diagnosis of gastrointestinal conditions relies a lot on image-based investigations and procedures (endoscopy and radiology). AI-assisted image analysis can make accurate assessment and provide more information than conventional analysis. AI integration of genomic, epigenetic, and metagenomic data may offer new classifications of gastrointestinal cancers and suggest optimal personalized treatments. In managing relapsing and remitting diseases such as inflammatory bowel disease, irritable bowel syndrome, and peptic ulcer bleeding, convoluted neural network may formulate models to predict disease outcome, enhancing treatment efficacy. AI and surgical robots can also assist surgeons in conducting gastrointestinal operations. While the advancement and new opportunities are exciting, the responsibility and liability issues of AI-assisted diagnosis and management need much deliberations.

The Tong Test: Evaluating Artificial General Intelligence Through Dynamic Embodied Physical and Social

摘要: The release of the generative pre-trained transformer (GPT) series has brought artificial general intelligence (AGI) to the forefront of the artificial intelligence (AI) field once again. However, the questions of how to define and evaluate AGI remain unclear. This perspective article proposes that the evaluation of AGI should be rooted in dynamic embodied physical and social interactions (DEPSI). More specifically, we propose five critical characteristics to be considered as AGI benchmarks and suggest the Tong test as an AGI evaluation system. The Tong test describes a value- and ability-oriented testing system that delineates five levels of AGI milestones through a virtual environment with DEPSI, allowing for infinite task generation. We contrast the Tong test with classical AI testing systems in terms of various aspects and propose a systematic evaluation system to promote standardized, quantitative, and objective benchmarks and evaluation of AGI.

Visible light responsive photocatalysts developed by substitution with metal cations aiming at artificial

摘要: To solve resource, energy, and environmental issues, development of sustainable clean energy system is strongly required. In recent years, hydrogen has been paid much attention to as a clean energy. Solar hydrogen production by water splitting using a photocatalyst as artificial photosynthesis is a promising method to solve these issues. Efficient utilization of visible light comprised of solar light is essential for practical use. Three strategies, i.e., doping, control of valence band, and formation of solid solution are often utilized as the useful methods to develop visible light responsive photocatalysts. This mini-review introduces the recent work on visible-light-driven photocatalysts developed by substitution with metal cations of those strategies.

Predicting the strength properties of slurry infiltrated fibrous concrete using artificial neural network

摘要: This paper is aimed at adapting Artificial Neural Networks (ANN) to predict the strength properties of SIFCON containing different minerals admixture. The investigations were done on 84 SIFCON mixes, and specimens were cast and tested after 28 days curing. The obtained experimental data are trained using ANN which consists of 4 input parameters like Percentage of fiber (PF), Aspect Ratio (AR), Type of admixture (TA) and Percentage of admixture (PA). The corresponding output parameters are compressive strength, tensile strength and flexural strength. The predicted values obtained using ANN show a good correlation between the experimental data. The performance of the 4-14-3 architecture was better than other architectures. It is concluded that ANN is a highly powerful tool suitable for assessing the strength characteristics of SIFCON.
